[A monosaccharide composition determination is the first step in the process of characterizing complex carbohydrate. Two frequently used methods, gas chromatography and high-performance liquid chromatography, will be discussed. Usually, the second step in carbohydrate structure characterization is the determination of the linkage types of the constituting monosaccharides. These assignments are obtained by gas–liquid chromatography-mass spectrometry. This chapter provides detailed protocols for monosaccharide and linkage analysis, as well as the absolute configuration (d/l) determination.]
