[This chapter will highlight techniques for the analysis of released glycans in their native, reduced, or labeled form. Nowadays, standardized chromatographic separations followed by mass spectrometric analysis and retention time comparison using on-line databases are the usual methods. Minimal amounts of material can be investigated due to fluorescent/chromophore labeling and the increased sensitivity of the analytical equipment. Fractionation and enzymatic sequence analysis will be discussed, in particular for N-glycans. The release and fractionation of O-glycans, as abundant components of mucins, will also be discussed. Extensive literature references are provided.]
